Cam chain timing

During the rebuild of my bike I installed a new cam chain, rings and valve job. Will the bike run with the timing off a tooth ? It seems sluggish and has a low end miss/hesitation and running rich and blackening the plugs. Could all of this stem from cam timing being off ? I have gone over the carb several times and the carb seems clean and correct jets are installed. Can I use a timing light to check ignition timing to see if I can match the F mark on the flywheel ?

The engine will also run HOT if a tooth off.
A timing light will help you decide to pull the engine and get the chain correct. .................lm

If I was a off on the cam timing then the correct F ignition timing will not be able to be achieved using the timing light. Correct ?
